NPR: Even one minute getting out of your chair adds to your mental health.  Fidgeting helps kids focus, a single bout of exercise gives mental health benefits, and if you exercise regularly for 10-20 minutes at a moderate intensity gives significant benefit, especially for people with depression.  Just stretching is as helpful as jogging.

 

Recently, Dr. Evans and a team of 18 researchers and clinicians reported the results of a first-in-human, Phase I clinical trial of a novel gene therapy for osteoarthritis. The findings, published in Science Translational Medicine, demonstrate that the therapy is safe, achieved sustained expression of a therapeutic gene inside the joint and offered early evidence of clinical benefit. This could revolutionize the treatment of osteoarthritis,” says Dr. Evans, who directs the Musculoskeletal Gene Therapy Research lab at Mayo Clinic. In osteoarthritis, the cartilage that cushions the ends of bones—and sometimes the underlying bone itself—degenerates over time. It is a leading cause of disability, and notoriously difficult to treat. Any medications you inject into the affected joint will seep right back out in a few hours,” says Dr. Evans. As far as I know, gene therapy is the only reasonable way to overcome this pharmacologic barrier, and it’s a huge barrier.” By genetically modifying cells in the joint to produce their own pharmacy of anti-inflammatory molecules, Evans aims to engineer knees that are more resistant to arthritis.    An article by Nelson et al discusses in detail why curcumin is so problematic and so far basically useless in medicine. Mitochondria are no longer viewed solely as ATP- or metabolite-generating organelles but as key regulators of cellular signaling that shape physiologic aging. Contrary to earlier theories linking aging to mitochondrial DNA mutations and oxidative damage, current evidence shows that these factors do not causally limit physiologic aging. Instead, an evolving literature links age-related loss of mitochondrial signaling and function to important physiologic changes of aging. Moreover, mild inhibition of mitochondrial respiratory function with drugs like metformin promote health span.

 

Exercise Identity and Spiritual Well-Being in Nursing Home Residents: Mediating Roles of Social Support and Exercise Behavior in Hebei Province, China.  Perceived social support and exercise behavior have parallel mediating effects on the relationship between exercise identity and Spiritual well-being of older adults residing in nursing homes. These findings offer a theoretical foundation for developing targeted interventions aimed at enhancing spiritual well-being through improved exercise identity and supportive environments in institutional care settings.

 

The Mediating Role of Activities of Daily Living in the Relationship Between Depressive Symptoms and Multidimensional Frailty in the Elderly with Multimorbidity. The findings confirm the importance of interventions targeting mental health and restoration of physical function to prevent multidimensional frailty in older adults with multimorbidity.

Oral Health and Diabetic Cardiomyopathy: Mechanisms, Biomarkers, and Early Screening Approaches. Diabetic Cardiomyopathy (DCM) is a common cardiovascular complication in patients with diabetes. In recent years, the association between oral health and diabetic heart disease has gained increasing attention. This perspective reviews the potential mechanisms of oral diseases in diabetic heart disease, oral indicators for early screening of diabetic heart disease, and proposes future research directions. The potential mechanisms of oral diseases in diabetic heart disease primarily involve abnormal activation of inflammatory responses, dysregulation of the oral microbiome, and immune system disorders. In the context of early screening for diabetic heart disease, oral health indices, salivary biomarkers, and the oral microbiome serve as critical oral indicators with significant clinical value for early diagnosis.

From Microbial Homeostasis to Systemic Pathogenesis: A Narrative Review on Gut Flora’s Role in Neuropsychiatric, Metabolic, and Cancer Disorders. Numerous studies have shown that curcumin can counteract the inflammatory response in sepsis and protect against organ damage caused by sepsis, suggesting that curcumin may become a new direction for sepsis treatment.

Atherosclerosis and Inflammation: Are the Rules of the Game Changing with Biological Therapies? Atherosclerosis is a chronic, progressive vascular disease characterized not only by lipid accumulation but also by sustained inflammation. Recent evidence has highlighted the critical role of immune pathways and pro-inflammatory cytokines in plaque formation, progression, and destabilization.

 

The Role of Uridine in Health and Disease. Since most tissues are unable to synthesize uridine independently, they rely on the circulatory system to uptake exogenous uridine to maintain basal cellular functions.  The continuous and stable circulating uridine level is the basis of the normal operation of various biological processes of the body, and the destruction of uridine homeostasis is bound to affect this, and then lead to disease. Obesity is a metabolic disease caused by excessive fat accumulation. It has been shown that the rhythm of uridine after meals is disrupted in mice fed with a high-fat diet or that are obese, and uridine supplementation can relieve abnormal uridine levels in states of obesity or high-fat diets, thereby mitigating obesity.  Osteoarthritis is a joint degenerative disease with an extremely high incidence in the elderly, and inflammation plays an important role in the manifestation of clinical events in osteoarthritis.79 Specifically, certain cytokines exhibit proinflammatory properties that are clearly activated during the course of the disease and significantly alter the homeostasis of the joint environment.80 A previous study reported9 that uridine improved osteoarthritis by reducing the aging of chondrocytes and mesenchymal stem cells.  The optimal dosage of UMP can vary depending on individual needs and health conditions. However, a common starting dose is typically between 150 mg to 250 mg per day. However sublingual doses have been documented to be 7-10 times more effective than processed through digestion. It is advisable to start with a lower dose and gradually increase it to assess tolerance and effectiveness.

 

Association of Recreational Physical Activity with Gut Microbiota Diversity and Composition, and Symptom Burden in Crohn’s Disease (CD): A Cross-Sectional Study. Promoting the incorporation of recreational physical activity (PA) as an indicator of healthy lifestyle management for CD may improve patients’ subjective psychological and mental well-being, while also preserving intestinal microbial diversity. Recreational PA is associated with favorable patient-reported outcomes and increased diversity of gut microbiota in CD, supporting its inclusion in lifestyle management strategies.

 

Gut Microbiome Composition Changes During Insomnia Treatment with Lemborexant. Our findings suggest that prolonged lemborexant treatment in individuals with insomnia may induce notable shifts in gut microbiota composition, including a significant reduction in Parabacteroides underscoring the potential interaction between hypnotic use and gut microbial balance.
