More who take Metformin
Oct 2021: Metformin is a drug that should be taken constantly, and twice a day in order to obtain the optimal benefit. The TAME study is at 850mg, twice a day. Adequate hydration, hepatic and renal function must be carefully maintained. It has serious side-effects, some fatal. It should not to be self-medicated. It must be adequately supervised by endocrinologist physician, who has great experience with this drug. He does not agree with the misinformation that metformin inhibits, or interferes with muscle growth and development. New York Times has article on harmful effect on muscles

Metformin approved 60 yrs ago in England for diabetes
Commonly used for lifetime, diabetics mortality lower than non-diabetics, Inhibits dementia, diabetes, covid, against many age-related diseases. In mice, extend 10% life, others 7-15%; similar to rapamycin. Intervention Testing Program at NIH-test drugs at 3 centers; metformin had no statistical effect-7%, 12%, -10% at three centers. Metformin is very cheap. Nir director of TAME
CAL Restriction (CALRES) – MET is CALRES mimetic; CALRES incr max lifespan; MET incr only healthspan, not lifespan; High dose MET is hazardous and shortens lifespan
Met does 2 things that fix aging. Energetic switch in mitochondria, but also in cells without mitochondria; transcripts change; glucose homeostasis-metformin mainly helps stabilize insulin level, makes beta-cells younger, increases insulin sensitivity; lowers glucose level; effects microbiome; perhaps not best orally, should be intravenous; approved for obesity treatment; MET transports glucose through kidneys for excretion if glucose elevated;

when started in late middle age. Open access: https://onlinelibrary.wiley.com/doi/full/10.1111/acel.13277
From Ken D: There have been questions raised about the effects of rapamycin on glucose. I’ve been taking rapamycin for several years now (5 mg per week). I also wear a CGM. In addition to rapamycin, I take 1000 mg of metformin daily. As you can see from the chart below I don’t have any glucose problems arising from my use of rapamycin. I should also mention that I stick to a low carb diet.
