Sci 28 Oct, pg 370-Spermidine decreases with age, supplementation in mature people activates aged T-cells and improves anti-tumor immunity. (I am now taking 10mg spermidine daily as a supplement).
The results demonstrated a clear correlation between the intake of spermidine and the improvement in cognitive performance in subjects with mild and moderate dementia in the group treated with the higher spermidine dosage.
Coffee intake of up to 5 cups per day was associated with significant reductions in the risks of cardiovascular disease, congestive cardiac failure, coronary heart disease, ischemic stroke, and various arrhythmias.

Handgrip strength and risk of cognitive outcomes: new prospective study and meta-analysis of 16 observational cohort studies. Handgrip strength (HGS), a measure of muscular strength, might be a risk indicator for cognitive functioning, but the evidence is not consistent. Using a new prospective study and meta-analysis of published observational cohort studies, we aimed to evaluate the prospective associations of HGS with poor cognitive outcomes including cognitive impairment, dementia and Alzheimer’s disease (AD). Meta-analysis of aggregate prospective data suggests that HGS may be a risk indicator for poor cognitive outcomes such as cognitive impairment, dementia and AD. Systematic review registration.
Variation in Gut Microbiota Composition is Associated with Sleep Quality and Cognitive Performance in Older Adults with Insomnia. Insomnia, a chronic condition affecting 50% of older adults, is often accompanied by cognitive decline. We tested associations between sleep quality and cognitive performance with gut microbiota in older adults with insomnia. Findings demonstrate the associations of sleep quality and cognitive performance with variance in gut microbiota composition and with specific genus abundance in older adults with insomnia.
Melatonin’s Benefits and Risks as a Therapy for Sleep Disturbances in the Elderly: Current Insights. Aging is accompanied by circadian changes, including disruptive alterations in the sleep/wake cycle, as well as the beginning of low-degree inflammation (“inflammaging”), a scenario that leads to several chronic illnesses, including cancer, and metabolic, cardiovascular, and neurological dysfunctions. Every day, the late afternoon/nocturnal surge in pineal melatonin helps to synchronize both the central circadian pacemaker found in the hypothalamic suprachiasmatic nuclei (SCN) and a plethora of peripheral cellular circadian clocks. Melatonin is an example of an endogenous chronobiotic substance that can influence the timing and amplitude of circadian rhythms. Moreover, melatonin is also an excellent anti-inflammatory agent, buffering free radicals, down-regulating proinflammatory cytokines, and reducing insulin resistance, among other things. We present both scientific and clinical evidence that melatonin is a safe drug for treating sleep disturbances in the elderly.

Earlier this year, a study from Korea suggested that IV vitamin C should be administered for at least five days to provide a significant benefit to sepsis patients. Since many randomized clinical trials only provided IV vitamin C for up to four days, extending the treatment period is one avenue to explore in future studies.
Senescent macrophages in the human adipose tissue as a source of inflammaging. Obesity is a major risk factor for type 2 diabetes and a trigger of chronic and systemic inflammation. Recent evidence suggests that an increased burden of senescent cells (SCs) in the adipose tissue of obese/diabetic animal models might underlie such pro-inflammatory phenotype. In morbid obesity, expansion of visceral adipose depots involves an increased burden of macrophages with senescent-like phenotype that may promote a pro-inflammatory profile and impair insulin signaling in adipocytes, supporting a framework where senescent macrophages fuel obesity-induced systemic inflammation and possibly contribute to the development of insulin resistance.
Mitochondrial ROS signalling requires uninterrupted electron flow and is lost during ageing in flies. Mitochondrial reactive oxygen species (mtROS) are cellular messengers essential for cellular homeostasis. In response to stress, reverse electron transport (RET) through respiratory complex I generates high levels of mtROS. Suppression of ROS production via RET (ROS-RET) reduces survival under stress, while activation of ROS-RET extends lifespan in basal conditions. Our results elucidate the mechanism by which ROS signalling is lost during ageing.
